Abstract The ß-carboline-1,3,5-triazine hydrochlorides 8-13 were evaluated in vitro against acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). The analysed compounds were selective to BuChE, with IC50 values in the range from 1.0-18.8 µM being obtained. The N-{2-[(4,6-dihydrazinyl-1,3,5-triazin-2-yl)amino]ethyl}-1-phenyl-ß-carboline-3-carboxamide (12) was the most potent compound and kinetic studies indicate that it acts as a competitive inhibitor of BuChE. Molecular docking studies show that 12 strongly interacts with the residues of His438 (residue of the catalytic triad) and Trp82 (residue of catalytic anionic site), confirming that this compound competes with the same binding site of the butyrylthiocholine

In the Myrtaceae family, the species Eugenia involucrata DC., popularly known as "cerejeira-do-mato", is traditionally used for the antidiarrheal and digestive action of its leaves. However, no studies were found in the literature regarding its antimicrobial and antioxidant potential. In this context, the objective of the present study was to determine the chemical composition by gas chromatography coupled to mass spectrometry (GC-MS) to evaluate the antimicrobial activity by the broth microdilution technique and the antioxidant activity by the 2,2-diphenyl-1-picryl-hydrazila (DPPH) method of the essential oil of E. involucrataleaves. GC-MS identified 28 compounds, all sesquiterpenes, corresponding to 89.41% of the essential oil. The antimicrobial activity of the essential oil was observed for all Gram-positive bacteria tested (Staplylococcus epidermidis, Enterococcus faecalis,Bacillus subtilis and Staplylococcus aureus) and for yeast Candida albicans. The essential oil presented a reduction capacity of DPPH up to 66.81%, evidencing its antioxidant potential. It is suggested that the antimicrobial and antioxidant action of E. involucrata essential oil is related to the presence of the major compounds, elixene (26.53%), ß-caryophyllene (13.16%), α-copaene (8.41%) and germacrene D (7.17%).


Na família Myrtaceae, a espécieEugenia involucrata DC. popularmente denominada "cerejeira-do-mato" é conhecida tradicionalmente pela ação antidiarreica e digestiva de suas folhas. Contudo, na literatura não foram encontrados trabalhos referentes ao seu potencial antimicrobiano e antioxidante. Neste contexto, o objetivo do presente estudo foi determinar a composição química por cromatografia gasosa acoplada a espectrometria de massas (CG-EM) e avaliar a atividade antimicrobiana pela técnica de microdiluição em caldo e a atividade antioxidante pelo método do 2,2-difenil-1-picril-hidrazila (DPPH) do óleo essencial das folhas de E. involucrata. A CG-EM identificou 28 compostos, todos sesquiterpenos, correspondendo a 89,41% do óleo essencial. A atividade antimicrobiana do óleo essencial foi observada para todas as bactérias Gram-positivas testadas (Staplylococcus epidermidis, Enterococcus faecalis,Bacillus subtilise Staplylococcus aureus) e para a levedura Candida albicans. O óleo essencial apresentou capacidade redutora de radicais DPPH de até 66,81%, evidenciando sua potencialidade antioxidante. Sugere-se que a ação antimicrobiana e antioxidante do óleo essencial de E. involucrata esteja relacionada à presença dos compostos majoritários, elixeno (26,53%), ß-cariofileno (13,16%), -copaeno (8,41%) e germacreno D (7,17%).

ABSTRACT Purpose: To establish whether the citrate concentration in the seminal fluid ([CITRATE]) measured by means of high-resolution nuclear magnetic resonance spectroscopy (1HNMRS) is superior to the serum prostate-specific antigen (PSA) concentration in detecting of clinically significant prostate cancer (csPCa) in men with persistently elevated PSA. Materials and Methods: The group of patients consisted of 31 consecutively seen men with histological diagnosis of clinically localized csPCa. The control group consisted of 28 men under long-term follow-up (mean of 8.7 ± 3.0 years) for benign prostate hyperplasia (BPH), with persistently elevated PSA (above 4 ng/mL) and several prostate biopsies negative for cancer (mean of 2.7 ± 1.3 biopsies per control). Samples of blood and seminal fluid (by masturbation) for measurement of PSA and citrate concentration, respectively, were collected from patients and controls. Citrate concentration in the seminal fluid ([CITRATE]) was determined by means of 1HNMRS. The capacities of PSA and [CITRATE] to predict csPCa were compared by means of univariate analysis and receiver operating characteristic (ROC) curves. Results: Median [CITRATE] was significantly lower among patients with csPCa compared to controls (3.93 mM/l vs. 15.53 mM/l). There was no significant difference in mean PSA between patients and controls (9.42 ng/mL vs. 8.57 ng/mL). The accuracy of [CITRATE] for detecting csPCa was significantly superior compared to PSA (74.8% vs. 54.8%). Conclusion: Measurement of [CITRATE] by means of 1HNMRS is superior to PSA for early detection of csPCa in men with elevated PSA.

Essential oils have aroused interest in the industrial sector due to the multitude of potential applications, especially with respect their antimicrobial and antioxidant properties, among others. The objective of this study was to determine the chemical composition of the essential oil from the leaves of Myrcia oblongata DC by gas chromatography coupled to mass spectrometry (GC-MS). To evaluate the antioxidant potential of the oil by using the free radical capture method with 2,2-diphenyl-1-picryl hydrazyl (DPPH); to test the oil antimicrobial activity using the broth microdilution method; and to evaluate the repellency and fumigant potential of the oil on Dermanyssus gallinae (Degeer, 1778). The GC-MS analysis resulted in the identification of 30 oil constituents, with the bulk of the composition identified as caryophyllene oxide (22.03%) and trans-verbenol (11.94%). The oil presented moderate antioxidant activity compared to the synthetic antioxidant 2,6-di-tert-butyl-4-hydroxytoluene (BHT). Antimicrobial activity of the essential oil showed an inhibitory activity on Gram-positive bacteria, Enterococcus faecalis, Staphylococcus aureus, Bacillus subtilis and Staphylococcus epidermidis and for the yeast Candida albicans, and showed no activity against Gram-negative bacteria. All concentrations of the essential oil used in the fumigation test on D. gallinae resulted in mortality below 20%. For the repellency test, significant potential was observed for the concentration of 10%.


Os óleos essenciais tem despertado interesse no setor industrial por apresentarem multiplicidade de aplicações, destacando-se por suas propriedades antimicrobianas, antioxidantes dentre outras. Diante disto, o objetivo deste estudo foi determinar a composição química do óleo essencial das folhas de Myrcia oblongata DC pelo método de cromatografia gasosa acoplada à espectrometria de massas (CG-EM), avaliar o potencial antioxidante do óleo pelo método de captura do radical livre 2,2- difenil-1-picril hidrazil (DPPH), testar a atividade antimicrobiana pelo método de microdiluição em caldo, bem como avaliar a repelência e potencial fumigante deste óleo sobre Dermanyssus gallinae (Degeer, 1778). As análises de CG-EM resultaram na identificação de 30 constituintes, sendo majoritários o óxido de cariofileno (22.03%) e o trans-verbenol (11.94%). O óleo apresentou atividade antioxidante moderada quando comparada ao antioxidante sintético 2,6-di-tert-butil-4-hidroxitolueno (BHT). Em relação à atividade antimicrobiana, o óleo essencial demonstrou atividade inibitória para as bactérias Gram-positivas, Enterococcus faecalis, Staphylococcus aureus, Bacillus subtilis e Staphylococcus epidermidis, e para levedura Candida albicans e não apresentou atividade para bactérias Gram negativas. Todas as concentrações do óleo essencial testadas no teste de fumigação sobre D. gallinae apresentaram mortalidade inferior a 20%. Para o teste de repelência, observou-se um potencial significativo para a concentração de 10%.

A series of novel hybrids ß-carboline-4-thiazolidinones were synthesized and evaluated for their in vitro antitumor activity against human cancer cell lines and for antiviral activity towards Herpes simplex virus type-1 (HSV-1). From the N'-(2-ylidene-4-thiazolidinone)-ß-carboline-3-carbohydrazide series (9-11), compounds 9c and 11d were the most active, showing growth inhibition 50% (GI50) values less than 5 µM for all cell lines tested. Compound 9c, bearing the 4-dimethylaminophenyl group at C-1 of ß-carboline was selected for further investigation concerning cell death and cell cycle profile, focusing on the human renal adenocarcinoma cell line 786-0. Treatments with 25 µM of compound 9c induced cell death after 15 h of treatment, characterized by phosphatidylserine exposure and loss of membrane integrity. Moreover, treatment with 12.5 µM promoted a sub-G1 arrest, which indicates cell death. Derivatives of the N-(2-substituted-aryl-4-thiazolidinone)-ß-carboline-3-carboxamide series (18-23) showed a potent activity and high selectivity for glioma (U251) and ovarian (OVCAR-3) cancer cell lines. Also, some ß-carboline-4-thiazolidinone hybrids showed potent antiviral activity against Herpes simplex virus type-1. The N-(2-substituted-aryl-4-thiazolidinone)-carboxamide moiety in 18, 19 and 22 confer a potent anti-HSV-1 activity for these derivatives, which presented EC50 values of 0.80, 2.15 and 2.02 µM, respectively. The assay results showed that the nature of 4-thiazolidinone moiety and of the substituent attached at the 3- and 1- position of ß-carboline nucleus influenced the antitumor and antiviral activities.

The methanol crude extract of the leaves of Ficus radicans Roxb. 'Variegata' (Moraceae) and the n-hexane, ethyl acetate and aqueous methanol fractions resulting from its fractionation were evaluated for their anti-inflammatory, molluscicidal and free-radical scavenging activities. The crude extract and fractions exhibited significant inhibition of inflammation in both croton oil (CO)-induced ear oedema in mice (p<0.001) and carrageenan-induced rat paw oedema models (p<0.01). The molluscicidal assay against Biomphalaria glabrata showed a weak activity for the n-hexane fraction (DL(50)= 400 µg mL(-1)). A moderated 1,1-diphenyl-1-picrylhydrazyl (DPPH) free-radical scavenging activity was observed for the ethyl acetate fraction (IC(50)= 66.2 µg mL(-1)). Fractionation of the extracts through chromatographic methods afforded the coumarins 7-methoxycoumarin, 7-hydroxy-6-methoxycoumarin and methoxy-3,4-dihydrocoumarin, the steroids ß-sitosterol and ß-sitosterol 3-O-ß-glucopyranoside, as well as a cinnamic acid derivative and a flavonoid identified as trans-4-methoxy-2-ß-D-glucopyranosyloxy cinnamic acid and quercetin 3-O-ß-D-xylopyranosyl-(1 → 2)-α-L-rhamnopyranoside, respectively. The compounds were identified on the basis of their NMR spectral data and comparison with those previously reported in the literature.

Objetivou-se com este estudo avaliar o potencial alelopático de extratos metanólicos de folhas, flores e raízes de capuchinha (Tropaeolum majus L.) sobre a germinação de sementes e o crescimento inicial de plântulas de picão-preto. O extrato metanólico com melhor potencial de inibição foi submetido a particionamento, resultando nas frações hexânica, clorofórmica, acetato de etila e hidrometanólica e posterior caracterização pelo espectro de absorção na região do infravermelho (IV). O efeito alelopático foi avaliado sobre as sementes de picão-preto, as quais foram distribuídas sobre papel germitest umedecido com 2mL dos extratos e mantidas em germinador do tipo B.O.D. regulado a temperatura de 25°C e luz branca constante, sendo que as sementes imersas diretamente em água constituíram o tratamento controle. A avaliação da qualidade da semente foi realizada pelos testes de germinação e vigor (primeira contagem e comprimento de raiz primária e de hipocótilo das plântulas), em delineamento inteiramente ao acaso. O potencial alelopático das folhas de capuchinha foi maior em relação às demais partes da planta sobre a germinação das sementes, comprimento de hipocótilo e de raiz das plântulas de picão-preto. Estes efeitos podem estar associados à presença de grupos químicos polares, pois à medida que se aumentou a polaridade dos solventes detectou-se maior efeito inibitório sobre a germinação e o crescimento inicial de plântulas de picão-preto.


This research aimed to evaluate the metanolic extracts allelopathic potential from leaves, flowers and roots of capuchinha (Tropaeolum majus L.) on picão-preto seeds germination and initial seedling growth. The best inhibitor metanolic extract was fractioned, in hexanic, cloroformic, etil acetate and hidrometanolic fractions and it was characterized through absorption spectrum using mid-infrared. To evaluate the allelopathic effect of metanolic extracts and the fractions aftereffect; it was used picão-preto seeds distributed on 2ml extracts moistured paper maintained in B.O.D. germinator at 25°C and white constant light. Seeds sowed directly in water moistured germitest paper constituted the control. The allelopathic potential extracts were evaluated through the first count, final germination percentage, seedlings primary root and hypocotyl length (cm). The experiment was carried out DIC with three replications of 25 seeds. The capuchinha leaves showed allelopathic potential on picão-preto seeds germination, hypocotyl and seedlings primary root length. It was suggested that such effects are associated to the presence of polar chemical groups because the increase in the solvents polarity detected the major picão-preto germination and seedlings growth inhibition effect.
